A total of 417 anchor microsatellite markers, used to construct a consensus genetic linkage map (Bihoreau et al. Genome Res 7: 434-440,1997), were selected to screen a 10-fold redundant YAC library (Li Cai et al. Genomics 39:385-392,1997). All YAC DNA pools were amplified by PCR using a single touchdown protocol ramping from 55C to 50C for the annealing step. One or more unambiguous YACs were identified for 261 anonymous markers and for 111 markers derived from genes. The YAC-STS framework provides at least one informative YAC clone every 5.4cM on average.
